We believe for most proteins, there are a certain number of regions could be promising antigens to produce high-quality antibodies. To maximize the chance of success, we usually apply a number of antigens (up to 20 protein fragments and/or peptides) instead of just one or two; the diverse antigens could map all the best possible regions of the target protein.
Based on our data from more than two thousand projects, in average, successful rate of applying 1 antigen is only 30%, whereas for 6 antigens is nearly 60%, and 10 antigens 70%. That is a clear demonstration of the power of multiplicity! This strategy is especially helpful for the “difficult” proteins like multi-pass transmembrane proteins.
